About this test
The sperm DNA fragmentation test measures damage to the genetic material (DNA) inside sperm, reported as the DNA Fragmentation Index (DFI). It reveals problems that a routine semen analysis may miss.
DFI interpretation:
- Below 15%: normal
- 15–30%: moderate
- Above 30%: high
High DNA fragmentation reduces fertility and is linked to poor embryo quality, IVF or IUI failure, and recurrent miscarriage. Causes include varicocele, smoking, infections, oxidative stress, heat exposure, obesity, and older age.

When should I get the SPERM DNA FRAGMENTATION test?
You should get a sperm DNA fragmentation test if you have unexplained infertility despite a normal semen analysis, recurrent miscarriages, or repeated IVF or IUI failure. It is also recommended for men with varicocele, smoking habits, obesity, exposure to heat or toxins, older age, or a history of genital infections or chemotherapy.
How should I prepare for the SPERM DNA FRAGMENTATION test?
No fasting is required for a sperm DNA fragmentation test. Abstain from ejaculation for 2–5 days beforehand. Avoid alcohol, smoking, hot baths, and saunas for a few days. Collect the sample by masturbation in a sterile container, without lubricants, and deliver it to the lab within 30–60 minutes. Inform your doctor about any recent fever or medicines.
Do I require fasting for the SPERM DNA FRAGMENTATION test?
No, fasting is not required for a sperm DNA fragmentation test. You can eat and drink normally before giving your sample. However, you should abstain from ejaculation for 2–5 days beforehand, avoid alcohol and smoking, and collect the sample without lubricants for accurate results.
